Inc bation of potato t ber tiss e discs on B5 medi m s pplemented with 1-naphtylacetic acid (NAA) led to call s formation, irrespective of the presence of kinetin ; witho t NAA no call s formation occ rred . Inc bation in the presence of abscisic acid (ABA) red ced the increases in fresh weight and dry weight both in call s-forming and in non-call s-forming tiss e. Mitochondrial respiration was lowered by ABA as well . The ind ction of the alternative, CN-resistant pathway was inhibited by the presence of ABA, especially in mitochondria from non-call s-forming tiss e .The in vivo respiration of the call s-forming tiss e was higher than that of the non-call sforming tiss e . Total respiration, cytochrome pathway activity and the capacity of the alternative pathway were all lowered in call s-forming tiss e by treatment with ABA . The in vivo activity of the alternative pathway was low in all tiss e types, especially after ABA-treatment . The slight stim lation by hydroxamates of the oxygen ptake of call s-forming tiss e inc bated on medi m with NAA and ABA indicates the involvement of a hydroxamateactivated peroxidase in the oxygen ptake of this tiss e ; this peroxidase seemed not to participate in the oxygen ptake of the other tiss es types .In non-call s-forming tiss e the oxygen ptake of ABA-treated tiss e was very low and almost completely resistant to the combined addition of inhibitors of both the cytochrome and the alternative pathway, indicating that the in vivo activity of the mitochondria in the oxygen ptake of the tiss e was very low . The possible ca ses for this ABA-effect are disc ssed . In non-call s-forming tiss e the treatment with ABA creates a sit ation which is comparable with that observed in intact potato t bers . This sit ation is characterized by a tiss e respiration lower than that of the isolated mitochondria and an alternative pathway capacity that is low or absent.

Pre-and postclimacteric Super Red Delicious apple (Malus domestica Borkh.) fruit were treated with ethephon, aminoethoxyvinylglycine (AVG), and diazocyclopentadiene (DACP) after harvest. Ethylene and volatile production were measured 1, 5, and 13 days after treatment. Production of some esters by preclimacteric fruit was enhanced after ethephon treatment, but the same treatment reduced production of various esters by postclimacteric fruit. Ethylene and volatile production were inhibited by AVG and DACP in preclimacteric fruit. Volatile acetate esters were not affected by any treatment, whereas AVG and DACP inhibited production of other volatile esters in postclimacteric fruits. The results suggest synthesis of some volatile esters requires continuous ethylene action and a high rate of ethylene production.

Three cytoplasmic male-sterile Nicotiana cultivars together with corresponding male-fertile progenitors and restored lines were investigated in order to find possible correlations between respiratory characteristics and male sterility. Oxygen consumption measurements were performed on cells from suspension cultures as well as on mitochondria isolated from green leaves. Inhibitors, which have been reported to specifically block either the cytochrome (KCN) or the alternative (propyl gallate and salicylhydroxamic acid [SHAM] respiratory pathways, were used in order to measure the capacity and activity of the two pathways. One of the inhibitors, SHAM, was found unsuitable to measure the activity of the altemative pathway due to the lack of specificity of SHAM for this pathway. A great difference in the capacity of the alternative pathway was detected between the two types of cell materials tested. Mitochondria isolated from green leaves showed a capacity of the alternative pathway of 5 to 20% of total mitochondrial repiration, while the capacity of cells from suspension cultures generally ranged from 50 to 80%. In addition to this, in organello synthesis of mitochondrial proteins revealed differences between mitochondria isolated from green leaves and from cell suspensions. No correlation, however, could be found between respiratory characteristics and male sterility.
