The relationship between lethal mutation yield and intake of ethylnitrosourea (ENU) in Drosophila melanogaster

Abstract: To estimate the absorbed dose of N-ethyl-N-nitrosourea (ENU) ingested in Drosophila melanogaster, males were fed with sucrose solutions containing various concentrations of ENU plus 3H-labeled sucrose for 24 hr. Flies showed decreasing intakes with increase in ENU concentration when monitored by intake 3H radioactivity. Absorbed dose, D, per male can be estimated by the following formula: D = v . C, where v is intake volume per male of sucrose solution at concentration C. Estimating the v value for each ENU so… Show more

“…consume the same amount of food at each level of mutagen, and second the amount of food consumed may vary from experiment to experiment. Various attempts to quantitate mutagen consumption have been reported [Gollapudi et al, 1985;Ayaki et al, 1984;Yoshikawa et al, 1984;Thomp-The quantitation of mutagenic responses to multiple exposures of a mutagen in feeding studies with Drosophilu is complicated by two main factors. “…As an alternative to dyes, the medium can be supplemented with radioactively labeled substances such as [ 14 C]-choline, [ 3 H]-thymidine or [α-32P]-dCTP [ 57 , 58 , 59 , 60 , 61 , 62 , 63 ]. Similar to the methods using dyed medium, flies are fed on labeled medium for a certain period of time.…”
