The role of cullin 5-containing ubiquitin ligases

Abstract: The suppressor of cytokine signaling (SOCS) box consists of the BC box and the cullin 5 (Cul5) box, which interact with Elongin BC and Cul5, respectively. SOCS box-containing proteins have ubiquitin ligase activity mediated by the formation of a complex with the scaffold protein Cul5 and the RING domain protein Rbx2, and are thereby members of the cullin RING ligase superfamily. Cul5-type ubiquitin ligases have a variety of substrates that are targeted for polyubiquitination and proteasomal degradation. Here, … Show more

“…Thus, ATF4 is unlikely to play a role in LATS1 expression at the transcriptional level. Because LATS1 expression is tightly controlled by post-translational modifications [28], we sought to examine the role of ATF4 in this process. Treatment with the proteasome inhibitor MG132 resulted in a higher induction of LATS1 expression in ATF4-deficient than proficient HT1080 cells and MEFs in response to H 2 O 2 treatment (Figure 4B, 4C).…”

“…Most of the 18 members of the chordate-specific ASB (Ankyrin Repeat and SOCS Box) gene family encode a specificity subunit of an E3 ubiquitin ligase complex [39, 60], and half of these genes are preferentially expressed in SkM or, less frequently, in SkM and heart (Table S4). The ankyrin repeats of this subunit determine specific protein-protein interactions that direct ubiquitination with subsequent proteosomal degradation of the target.…”

“…4c). Since many CRL complexes are known to form dimers, and the dimerization is often mediated by the substrate binding or adaptor proteins, this dimer of the Vif complex may be the functionally relevant species (48)(49)(50)(51)(52)(53)(54)(55). We collected methyl-TROSY NMR of the 13 C-ILVMA-labeled VCBC complex bound to dT15 (Fig.…”
