The role of endogenous reactive oxygen species in oxymatrine-induced caspase-3-dependent apoptosis in human melanoma A375 cells

Abstract: Rapid increases in incidence and mortality of human malignant melanoma are observed worldwide; thus, the development of new effective chemicals to control melanoma is urgent. In this study, the cytotoxic effect of oxymatrine, a natural quinolizidine alkaloid, against three human melanoma cell lines (A375, Sk-Mel-28, MM96L) and the underlying mechanisms were investigated. Oxymatrine killed all three human melanoma cell lines in a dose-dependent manner. The compound also dose-dependently caused apoptosis in huma… Show more

“…Degradation of the ECM of blood or lymph vessels is critical to metastasis because loss of the ECM allows cancer cells to invade the blood or lymphatic system and spread to distal tissues and organs [20]. MMPs, especially MMP-2 and MMP-9, are believed to be potent in degrading ECM [7,17,21]. Matrine has been proved to be able to reduce the expression of MMPs in many cancers [22,23].…”

“…5 It triggers oxidative stress, results in the collapse of the mitochondrial transmembrane potential and in turn leads to cytochrome c release and apoptosis through the intrinsic caspase-9/caspase-3 pathway in melanoma cells. 6 Cancer stem cells (CSCs) play a critical role in cancer initiation and relapse as they are resistant to most cytotoxic agents and able to proliferate indefinitely. OMT inhibits CSC growth through inhibition of the Wnt/β-catenin signaling pathway, and represents a novel therapeutic drug for targeting breast CSCs.…”

“…Human melanoma cell lines, including WM35 (25), WM451 (25) and SK28 (26), HaCaT (27) and Hm (25) cells were purchased from the Chinese Academy of Sciences (Shanghai, China). Cells were cultured in Dulbecco's modified Eagle's medium (DMEM; HyClone, UK) with 10% fetal bovine serum (FBS; Gibco, USA) in an 37˚C atmosphere of 5% CO 2 .…”
