The Role of HLA-G in Tumor Escape: Manipulating the Phenotype and Function of Immune Cells

Abstract: Human leukocyte antigen-G (HLA-G) is a non-classical major histocompatibility complex class I (MHC I) molecule, and under physiological conditions, its expression is strictly restricted to the maternal–fetal interface and immune-privileged organs where HLA-G is expected to contribute to establishment and maintenance of immune tolerance. However, the expression of HLA-G has been found in various types of tumors, and the level of its expression frequently correlates with high-grade histology and poor prognosis, … Show more

“…Moreover, the observed phenotypes are similar to those of VEGF pathway mutants ( Carmeliet et al, 1996 ; Damert et al, 2002 ; Ferrara and De Vanna, 2016 ). Loss of Rreb1 also led to increased expression of H2-Q2, a homolog of HLA-G that regulates VEGF expression through indirect mechanisms ( Liu et al, 2020 ) as well as vasculogenesis and differentiation of blood lineages ( Comiskey et al, 2003 ; Liu et al, 2020 ). As Rreb1 functions downstream of, and regulates, receptor tyrosine kinase signaling, its role in vasculogenesis may be mediated via VEGF.…”

“…Thus, Rreb1 -/- cells might perform a role comparable to leader cells in cancer metastasis, remodeling the ECM to permit migration of wild-type neighbors. HLA-G (H2-Q2) upregulation is also associated with metastasis and immune cell evasion ( Liu et al, 2020 ) and, as a secreted factor, might also promote invasive behaviors in both wild-type and mutant cells.…”

“…In contrast to HLA-A,-B,-C and -E, HLA-G is expressed on a restricted set of cells and tissues and mainly in tissues characterized by a tolerogenic- or immune suppressive immune environment ( 171 ). Clear examples are the expression of HLA-G on trophoblast cells of the developing fetus during pregnancy or the enhanced expression of HLA-G on tumor or tumor-accessory cells contributing to immune evasion in cancer ( 172 , 173 ). Also in the solid organ transplantation setting, HLA-G induced tolerance for the allograft has been described: In brief, HLA-G contributes to short- term tolerance by interacting with inhibitory receptors, like LILRB1/2 and KIR2DL4 on immune effector cells.…”

“…Эти рецепторы связываются с HLA-G в 3-4 раза сильнее, чем с классическими MHCI, что указывает на ведущую роль такого взаимодействия в регуляции активности Т-клеток и антиген-презентирующих клеток. Кроме того, ILT2 и ILT4 рецепторы конкурируют с CD8+ лимфоцитами за связывание с MHCI, следствием чего является угнетение их цитотоксичности [34]. Такие особенности микроокружения опухоли могут отразиться на эффекте ИИТ.…”
