The Role of Nrf2 in the Attenuation of Cardiovascular Disease

Abstract: Oxidative stress is a component of many human diseases, including cardiovascular diseases (CVD). Exercise and various phytochemicals activate nuclear factor (erythroid-derived 2)-like 2 (Nrf2), the master regulator of antioxidant defenses, and attenuate CVD. This review highlights Nrf2 regulation by exercise and phytochemicals and the role of Nrf2 as a therapeutic target in CVD.

“…For some of the materials, studies were done to show activity of the mixture, but in other cases reliance was simply made on known properties of the individual ingredients. In at least one case, Protandim (LifeVantage, Inc, Sandy, UT, USA), the complete mixture has been extensively studied, not just the individual ingredients 40,186,191200. Study of the combined ingredients allowed demonstration of synergistic interaction between the ingredients; each of which separately was previously known to be a Nrf2 activator 191.…”

“…Recently, a relationship between exercise and Nrf2 activation has been demonstrated 200,202205. For example, activation of Nrf2 was induced by acute exercise in a mouse model, and exercise-induced oxidative stress was higher in Nrf2−/− mice due to lower expression of Nrf2-dependent antioxidant genes 204.…”

“…In the developed countries CVD is one of the leading causes of death. 18 Overproduction of oxidants ameliorates the severity of the disease. Oxidative damage results into endothelial cell injuries and deleterious vasodilator effects.…”

“…Further reduction reactions can subsequently give rise to hydrogen peroxide [ 9 , 10 ], which has long been thought of as a harmful molecule; however, recently, new evidence has emerged which suggests that at low concentrations hydrogen peroxide acts as an intracellular signaling molecule involved in survival and proliferation mechanism. In contrast, exogenous ROS is produced by many environmental mediators which have demonstrated involvement in a number of pathological states including cardiovascular disease [ 11 ], chronic inflammation [ 12 ], and neurodegenerative diseases [ 13 ] as well as cancer [ 14 ].…”
