The role of p38 mitogen‐activated protein kinase in IL‐6 and IL‐8 production from the TNF‐α‐ or IL‐1β‐stimulated rheumatoid synovial fibroblasts

Abstract: We examined the role of p38 mitogen-activated protein (MAP) kinase in the tumor necrosis factor(IL-6) and interleukin-8 (IL-8) in fresh rheumatoid synovial fibroblast (RSF) cultures concomitantly with the induction of p38 MAP kinase activity. Pretreatment of RSF with a specific p38 MAP kinase inhibitor, SB203580, blocked the induction of IL-6 and IL-8 without affecting nuclear translocation of nuclear factor U UB (NF-U UB) or IL-6 and IL-8 mRNA levels. These findings suggest that p38 MAP kinase inhibitor may h… Show more

“…Upon cellular stimulation, IκB-α protein is phosphorylated and targeted for ubiquitination and subsequent degradation by the 26S proteosome (Chang and Karin 2001). NF-κB subsequently is released and translocated to the nucleus to activate the expression of various target genes (Suzuki et al 2000;Wang et al 2003). Pro-inflammatory cytokines such as TNF-α and IL-1β can also activate NF-κB through signal transduction involving the TNF receptor associated factor (TRAF) family of adaptor proteins (Dempsey et al 2003).…”

“…As a matter of fact, p38 MAPK plays a major role in facilitating the cytokine activation induced by TNF. Induction of proinflammatory mediators such as IL-1 and IL-6 by TNF is mediated by activation of p38 MAPK (31). Of note, these cytokines are inducibly expressed in these initial stages of arthritis and appear to be selectively linked to the areas of onset of inflammation, such as the cellular infiltration of the tendon sheaths.…”

“…2 The p38 MAP kinase is of particular interest in inflammatory diseases such as rheumatoid arthritis (RA) because it regulates the production of pathogenic cytokines such as interleukin (IL)-1 and tumor necrosis factor (TNF)-␣. 3,4 p38 is expressed and activated in RA synovium 5 and blockade using selective inhibitors decreases inflammation and bone destruction animal models of arthritis. 6 However, little is known about the upstream kinases that can activate this pathway in joint tissues.…”
