The Role of S100A6 in Pancreatic Cancer Development and Its Clinical Implication as a Diagnostic Marker and Therapeutic Target

Abstract: Recent microarray analyses showed that the S100 family contains members that are candidate diagnostic markers or therapeutic targets. In the present study, to evaluate the involvement of S100A6 in pancreatic cancer and its clinical usefulness for diagnosis, we examined S100A6 mRNA expression in pancreatic tissues and pancreatic juice from patients with different pancreatic diseases. To investigate the role of S100A6 in carcinogenesis of pancreatic cancer and the potential of S100A6 as a diagnostic marker for e… Show more

“…29 Total RNA was isolated from cancer cell lines using the High Pure RNA Isolation Kit (Roche Diagnostics, Mannheim, Germany) with DNase I (Roche Diagnostics) treatment, in accordance with the manufacturer's instructions. RNA was quantified by measuring absorbance at 260 nm, and purity was evaluated from the 260/280 ratio of absorbance using a NanoDrop ND-1000 spectrophotometer (NanoDrop Technologies, Wilmington, DE, USA).…”

“…After microdissection, total RNA was extracted from the selected cells and subjected to real-time RT-PCR for quantitative measurement of REG4 and CDX2 mRNA as described previously. 26,27 Quantitative Assessment of REG4 and CDX2 mRNA Levels by One-Step qRT-PCR We designed specific primers as follows: REG4, 5 0 -CAGATCCTGGTCTGGCAAGT-3 0 (forward) and 5 0 -ATTCGTTGCTGCTCCAAGTT-3 0 (reverse); CDX2, 5 0 -CTCCTCCCCAGCTCTTCTCT-3 0 (forward) and 5 0 -CCT TTCCCTTGAGTCCCTCT-3 0 (reverse); 18S rRNA, 5 0 -GT AACCCGTTGAACCCCATT-3 0 (forward) and 5 0 -CCATC CAATCGGTAGTAGCG-3 0 (reverse). One-step qRT-PCR was performed using a QuantiTect SYBR Green RT-PCR kit (Qiagen) with a Chrom4 Real-Time PCR Detection System (Bio-Rad Laboratories, Hercules, CA, USA) as described previously.…”

“…Many members of the EF-hand calcium sensor family play an important role in regulating tumor cell invasiveness (21, 22). In some cases, the level of S100A6 expression correlates with high metastatic activity, suggesting its involvement in increased cell invasion (23). S100A4 has also been shown to regulate cell motility and invasion to promote cancer metastasis (24).…”
