The role of the myofibroblast in tumor stroma remodeling

Abstract: Since its first description in wound granulation tissue, the myofibroblast has been recognized to be a key actor in the epithelial-mesenchymal cross-talk that plays a crucial role in many physiological and pathological situations, such as regulation of prostate development, ventilation-perfusion in lung alveoli or organ fibrosis. The presence of myofibroblasts in the stroma reaction to epithelial tumors is well established and many data are accumulating which suggest that the stroma compartment is an active pa… Show more

“…S1C). These findings suggest that LOXL2 facilitates fibroblast activation and production of a strong fibrous ECM structure in 4T1 tumors, both of which are known to enhance tumor cell invasion and metastasis (32)(33)(34)(35)(36).…”

“…CAFs have been identified as potent stimulators of cancer metastasis (102,103). They contribute to remodeling of the tumor microenvironment and promote its stiffness mediating a mesenchymal phenotype in epithelial cells by loosing their cell-cell junctions and polarity (104,105). The generation of CAFs by tumor cellderived exosomes has been described in numerous examples.…”

“…The breach of the epithelial layer is followed by changes that occur in the underlying connective tissue resulting in the loss of tissue homeostasis [1] . Normal wound healing is a well-known phenomenon.…”

“…However, interferon-γ, a basic fibroblast growth factor (bFGF), prostaglandin E2, and high cell density inhibit the differentiation of protofibroblasts to MFs [20] . After activation, MFs initiate the synthesis of a new collagencontaining matrix that consists proteoglycans and glycosaminoglycans (highly hydrated molecules) [1,22,24] . They produce tractional forces at the margins for wound contraction, which is known as tractional remodelling [22] .…”
