The Ror-Family Receptors in Development, Tissue Regeneration and Age-Related Disease

Abstract: The Ror-family proteins, Ror1 and Ror2, act as receptors or co-receptors for Wnt5a and its related Wnt proteins to activate non-canonical Wnt signaling. Ror1 and/or Ror2-mediated signaling plays essential roles in regulating cell polarity, migration, proliferation and differentiation during developmental morphogenesis, tissue-/organo-genesis and regeneration of adult tissues following injury. Ror1 and Ror2 are expressed abundantly in developing tissues in an overlapping, yet distinct manner, and their expressi… Show more

“…Interestingly, both receptors share an evolutionary conserved FZD domain in the ECD ( Yan et al, 2014 ). ROR1 and its closely related paralog ROR2 are receptors for Wnt5a and other Wnts in the planar cell polarity pathway ( Endo et al, 2022 ; Green et al, 2014 ; Minami et al, 2010 ). MFRP is Type II transmembrane protein with an extracellular FZD domain.…”

“…Intestingly, both receptors share a evolutionary conserved Frizzled domain in the ECD (Yan et al, 2014). ROR1 and its closely related paralog ROR2 are receptors for Wnt5a and other Wnts in the planar cell polarity (PCP) pathway (Endo et al, 2022; Green et al, 2014; Minami et al, 2010). MFRP (Membrane frizzled-related protein) is type II transmembrane protein with an extracellular FZD domain.…”

“…Noncanonical WNT signals are less well studied than canonical signals. Wnt5a is one of the typical noncanonical WNT ligands and can bind to its receptor Ror2, which belongs to the family of tyrosine-protein kinase transmembrane receptors (83,84). Interestingly, deletion of Ror2 in the root progenitor cells of the mouse molar leads to a reduction in root length, suggesting noncanonical WNT signaling plays a vital role in tooth development (85).…”
