The TLR4‐associated phospholipase D1 activation is crucial for Der f 2‐induced IL‐13 production

Abstract: Our data extend the knowledge for a variety of possible roles of PLD1 in allergic disorders including asthma pathogenesis and suggest possible candidacy of PLD1 as a molecular target for novel therapeutic approaches.

“…showed some years ago that TLR4 signaling could be initiated by the major HDM allergen Der p 2, which mimics the lipopolysaccharide (LPS)‐binding subunit MD‐2, leading to the facilitation of TLR4 signaling. Similarly, HDM allergen Der f 2 can bind directly to TLR4 and elicit pro‐inflammatory cytokine production in human bronchial ECs …”

“…Similarly, HDM allergen Der f 2 can bind directly to TLR4 and elicit pro-inflammatory cytokine production in human bronchial ECs. 67 Since a substantial proportion of HDM allergens are proteases, attention has also been given to the involvement of protease-activated receptors (PARs) on ECs. Among the four PAR family members, only PAR-2 can be activated by serine proteases that are present in HDM.…”

“…Der p 2 activates the innate immune system through Toll-like receptors (TLR-4) by mimicking the action of human MD-2 (myeloid differentiation antigen-like lipid-binding protein), a structural homologue that loads lipopolysaccharide (LPS) onto these receptors [12]. A TLR4-associated phospholipase D1 activation has recently been reported to be crucial for Der f 2-induced IL-13 production [13]. …”

“…However, little is known about the signals released by allergens that result in the activation of these receptors. Recent report from Choi et al [8 ▪ ] identified a Toll-like receptor 4 (TLR4)-associated phospholipase D1 (PLD1) signaling cascade that was crucial for house dust mites (HDM)-induced production of IL-13. The finding not only sheds light on the mechanism of HDM-mediated allergic inflammation but also suggests PLD1 as a novel therapeutic target.…”
