The Structures of Pyridovericin and Pyridomacrolidin, New Metabolites from the Entomopathogenic Fungus, Beauveria bassiana.

Abstract: Entomopathogenic fungi seem to be interesting as a source of biologically active compounds such as an insecticides1) and immunosuppressants2,3), because all insect orders are susceptible to fungal diseases. With the goal of finding novel bioactive compounds from fungi, we investigated metabolite diversity using an HPLC screening program. During the screening process, we isolated two novel metabolites named pyridovericin (1) and pyridomacrolidin (2) (Fig.

“…In the course of an HPLC-based, chemical screening program of the metabolites of entomopathogenic fungi, the novel pyridone alkaloids, pyridovericin and pyridomacrolidin, possessing protein kinase inhibitory activity, were isolated from the cultured broth of Beauveria bassiana EPF-5 [8,9]. In addition, novel quinolone alkaloids, quinolactacins A (1), B (2), and C (3) were obtained from the cultured broth of Penicillium sp.…”

“…However, also certain non-pigmented strains, such as Paecilomyces cateniannulatus RCEF 0108 and Paecilomyces militaris RCEF 0261 showed pronounced activity in the PC-12 assay. It is possible that they may contain structurally related alkaloids which lack the conjugated side chain of militarinones A-D. Colourless pyridone alkaloids like pyridovericin and pyridomacrolidin have indeed been isolated from entomopathogenic fungi (Takahashi et al, 1998). The identification of the cytotoxic substances in Paecilomyces militaris RCEF 0261 extracts is another priority for future investigations.…”

“…Entomogenous fungi are an ecologically highly specialized group of microorganism, which produce a plenty of insecticidal and pharmaceutical active compounds (Isaka et al, 2005;Molnar et al, 2010;Namatame et al, 1999;Takahashi et al, 1998). Especially, the insect fungal secondary metabolite Myriocin (ISP-I) derived fingolimod (Gilenya; FTY720) was approved (September 2010) by the U.S. FDA as a new treatment for multiple sclerosis (Strader et al, 2011).…”
