The synthesis of human α‐2‐HS glycoprotein is down‐regulated by cytokines in hepatoma HepG2 cells

Abstract: The regulation of the synthesis of a-2-HS glyeoprotein (AHSG) by inflammatory mediators from activated monocytes was studied on the human hepatoma cell line HepG2 and compared to that of albumin. Monocyte-conditioned medium, recombinant human interleukin-6 (rhlL6) and interleukin-lfl (rhlLlfl) all down-regulated the synthesis of AHSG. This decrease was found both at the protein and the mRNA level. The most efficient mediator was the monocyte-conditioned medium, when rhlLlfl was found to be less efficient than … Show more

“…insulin levels and inflammation) could also influence the synthesis or degradation of fetuin-A. In fact, interleukin (IL)-6, IL-1 and tumor necrosis factor-(TNF-) downregulate fetuin-A synthesis 27,28) . Thus, serum fetuin-A levels in subjects with DM may vary among clinical settings.…”

“…Fetuin-A is known to induce proinflammatory cytokines, such as IL-6, IL-1 and TNF- 32) . On the other hand, fetuin-A is a negative acute phase protein and its expression is suppressed by these cytokines 27,28) . Thus, the cross-sectional correlation between fetuin-A and CRP is complex, and the association between fetuin-A and CRP may alter depending on the type, grade or phase of disease.…”

“…hyper/hypoinsulinemia and inflammation) could also influence AHSG gene transcription. In fact, it has been reported that interleukin-6, interleukin-1 24) and tumor necrosis factor- 25) downregulate AHSG synthesis. In addition, we did not examine the chronic effect of high glucose concentrations on AHSG transcription/expression, which may be different from the short-term effect.…”

“…25,26 The synthesis of human AHSG in the liver is downregulated by pro-inflammatory cytokines, indicating that this protein is a negative acute-phase reactant. 27 Estrogen might also influence the production of the protein, as suggested by the changes in its serum levels after menopause and estrogen therapy. 28 In addition, an impaired renal function was observed to be associated with decreased circulating AHSG levels, and the degree of glomerular dysfunction correlated with AHSG levels in patients with chronic kidney disease.…”
