The systemic use of procaine in the treatment of the elderly: a review

Abstract: This article is a review and evaluation of the world literature on the systemic use of procaine in the treatment of the aging process and the common chronic diseases of later life. Included are data from 285 articles and books, describing treatment in more than 100,000 patients in the past 25 years. Except for a possible antidepressant effect, there is no convincing evidence that procaine (or Gerovital, of which procaine is the major component) has any value in the treatment of disease in older patients. “…The beneficial effects of procaine on orientation, attention, memory, psychic depression, libido, appetite, euphoria and various types of pain, which were initially described by Aslan (1956) and subsequently confirmed by various authors (Ostfeld et al, 1977), could be explained by the improvement of the functioning of the cholinergic system.…”

“…The demonstration that procaine, lignocaine and bupivacaine induce analgesia through a central mechanism which is antagonized by atropine, and HC-3 throws new light on the controversial question of the systemic use of procaine in the treatment of the elderly. “…In addition to enhancing memory functions, this drug was touted as having posi tive effects on many disease conditions associated with aging. However, basic studies suggest that the usual dose of intramuscularly administered procaine does not reach a concentration sufficient for pharmacological effects ex cept at the injection site [Ostfeldcl al., 1977], Most wellcontrolled clinical studies demonstrate no beneficial ef fect of Gerovital on physical or mental symptoms with the possible exception of transient mood elevation [Jarvik and Milne, 1975;Ostfeld et al. 1977].…”

“…Procaine is a known local anesthetic drug that has been used in geriatric treatment [6]. Although it has been shown to retard rat skin wound healing [7], some experiments suggest that procaine could accelerate the wound repair process.…”
