The Transposon-Encoded Protein TnpB Processes Its Own mRNA into ωRNA for Guided Nuclease Activity

Abstract: TnpB is a member of the Obligate Mobile Element Guided Activity (OMEGA) RNA-guided nuclease family, is harbored in transposons, and likely functions to maintain the transposon in genomes. Previously, it was shown that TnpB cleaves double- and single-stranded DNA substrates in an RNA-guided manner, but the biogenesis of the TnpB ribonucleoprotein (RNP) complex is unknown. Using in vitro purified apo TnpB, we demonstrate the ability of TnpB to generate guide omegaRNA (ωRNA) from its own mR… Show more

“…3B). Similar results were recently described for other TnpB homologs, suggesting a generalizable role of transposon-encoded nucleases in ωRNA biogenesis (61). Whether a similar feature is also true for related bacterial IscB nucleases and eukaryotic Fanzor nucleases remains to be determined.…”

“…7 ). Our finding might be extended to diverse RNA-guided systems, including different TnpB nucleases 52 , 61 , 62 , Cas12 nucleases and recently identified eukaryotic Fanzor nucleases that are closely related to TnpB 31 33 , 63 .

“…TnpB and IscB nucleases process the ends of the transposon-encoded RNA transcript into ωRNA, which complex with the respective nucleases to form a RNA-guided dsDNA endonuclease ribonucleoprotein (RNP) ( 4 , 6 , 9 ). We searched Fanzor loci for putative regions encoding OMEGA-like RNAs, based on conservation of noncoding sequence.…”
