The use of BIOCHIP mosaics in diagnostics of bullous pemphigoid: Evaluation and comparison to conventional multistep procedures

Abstract: BackgroundBullous pemphigoid (BP) is an autoimmune blistering disease associated with autoantibodies against BP180 and/or BP230 antigens. The immunoassays available for serological diagnostics include indirect immunofluorescence (IIF) on monkey esophagus (ME), salt‐split skin (SSS), and enzyme‐linked immunosorbent assay (ELISA) for BP180‐NC16a and BP230. Only a few studies validated innovative BIOCHIP mosaic, but none compared agreement between BIOCHIP substrates with conventional methods separately.MethodsWe … Show more

“…[8][9][10][11][12]16,17 This is compared to sensitivities of 45.1-66.7% for the BP230 mosaic on the BIO-CHIP, and a specificity of 39.0-100%. [8][9][10][11][12]16,17 The lower sensitivity of the BP230 mosaic compared to the BP180 mosaic is not unexpected given that it is more common to have autoantibodies against BP180, compared to BP230 in bullous pemphigoid.…”

“…8,9 In bullous pemphigoid, the sensitivity of the BP180 mosaic on the BIOCHIP has been found to range more widely between 55.3 and 100%, [8][9][10][11][12]16,17 with a specificity between 96.5 and 100%. “…The results were measured by an ELISA plate reader (KHB-ST 360). The standard recommended cutoff values were 20 RU/ml for both BP-180 NC16a and BP-230 ( 9 ).…”

“…In our study, we used the BIOCHIP Dermatology Mosaic 7 (Euroimmun, Lubeck, Germany), which consists of (i) monkey esophagus (BIOCHIP-ME), (ii) salt split skin (BICHIP-SSS), (iii) recombinant BP180 NC16a, (iv) HEK293 cells transfected with BP 230, (v) HEK293 cells transfected with Dsg1, and (vi) HEK293 cells transfected with Dsg3. The assay was carried out in accordance with the manufacturer’s instructions ( 9 ).…”

“…It has broad application prospects, namely, clinical diagnosis, industrial control, food and drug analysis (including biopharmaceutical research and development) in the national economy, environmental protection, biotechnology, biochips, and other research areas. With the development of science and technology, biotechnology and electronics infiltrate and merge ulteriorly, therefore more and more electrochemical biosensors have been designed to solve the detection problem of ultra-low concentration [6][7][8].…”
