Therapeutic Duration and Extent Affect the Effect of Moxibustion on Depression-Like Behaviour in Rats via Regulating the Brain Tryptophan Transport and Metabolism

Abstract: Moxibustion has been widely accepted as an alternative therapy for major depressive disease (MDD). However, the efficacy of moxibustion treatment on MDD is highly variable because of its irregular operation. This study was designed to investigate how therapeutic duration and extent influence the anti-depression effect of moxibustion and the underlying mechanism involved. Rats with lipopolysaccharide-induced depression-like behavior were treated by moxibustion treatment. The anti-depression effect was determine… Show more

“…Another study on suspended moxibustion for focal cerebral ischemia/reperfusion injury indicated a greater anti-apoptotic effect of 35minute moxibustion than 15-minute moxibustion [13]. When treating depression-like behavior disorder in rats, prolonged duration of moxibustion under the optimal extent could increase the level of 5-HT [12]. In the treatment of chronic neck pain with the direct moxibustion of small moxa cone, a trend of effect improvement was related to an increase of the amount of moxa cones, when the other dose factors were constant [11].…”

“…A variety of factors in uence the therapeutic e cacy of moxibustion, especially moxibustion dose, which includes moxibustion duration, moxibustion frequency, moxibustion amount-size and number of moxa cone [11][12][13][14]. None studies have focused on the dose-effect of moxibustion for CFS up to now, therefore, we designed this trail to investigate the relationship of long-snake-like moxibustion duration and effect in the treatment of CFS by combining subjective scales and objective measurement for assessment.…”

“…As the interventions were not completely the same in the included studies, we divided the study into five subgroups according to the specific intervention methods and performed meta-analysis of the overall efficacy and the efficacy of the subgroups. Also, previous studies have shown that moxibustion works by promoting the absorption of Trp in the brain and transferring the metabolism of Trp to serotonin, which can increase the production of serotonin and exert an antidepressant effect (25). In addition, compared to Western drugs for PSD, the advantages of traditional Chinese medicine (TCM) were safety and low side effects.…”

“…Several studies have supported the anti-depression effect of moxibustion. Even more importantly, prolonged treatment of moxibustion could promote the brain uptake of L-tryptophan (Trp), shifting the Trp metabolism to 5-hydroxytryptamine (5-HT) (25)(26)(27).…”

“…A systematic review showed that moxibustion could participate in inhibiting oxidative stress and cell apoptosis, adjusting inflammation and Aβ genesis activation of vascular endothelial growth factor, and regulating the metabolic products in the tricarboxylic acid cycle and fatty acid metabolism (Aum et al, 2021 ). In addition, moxibustion could alleviate the depression-like behavior and adjust tryptophan transport and 5-HT generation (Li et al, 2019 ). Ha et al ( 2020 ) found that the effect of moxibustion on the cognition of aging mice is associated with the genes and proteins involved in the APP metabolic pathway.…”
