Therapeutic efficacy of magnesium after acoustic trauma caused by gunshot noise in guinea pigs

Abstract: The threshold shifts of the 170-dB SPL group that received magnesium were significantly lower than those of controls after 2 and 7 days of recovery, but no significant difference was evidenced at 14 days in this group, nor at any time in the 176-dB SPL group. Animals treated with magnesium after the 176-dB SPL exposure had a significant reduction in hair cell loss in the basal region.

“…To limit the individual variability, the methodology consisted of comparing the two ears of the same animal. Indeed, in previous experiments, we did not observe any significant differences in hair cell losses between the left and the right ear in the same animal exposed to impulse noise [7,8]. This methodology has been previously used in different studies [9,10].…”

“…Two days after gunshot noise exposure, z-VAD-FMK-treated ears displayed significantly lower hearing loss at high frequencies (8,16,19 and 22 kHz). On days 7 and 14, hearing loss was equivalent in treated and non-treated ears.…”

“…Threshold shift reduction 7 days after acoustic trauma in guinea pigs was confirmed using DPOAE (Haupt et al, 2003). In another study (Sendowski et al, 2006c) it has been shown that a 7-day post-trauma magnesium treatment reduced auditory threshold shift measured 7 days after gunshot noise exposure. However, this improvement was temporary, suggesting that it could be potentially beneficial to prolong magnesium administration.…”
