Therapeutic Targeting of Th17/Tc17 Cells Leads to Clinical Improvement of Lichen Planus

Abstract: Lichen planus (LP) is a common, chronic relapsing inflammatory disorder of the skin and mucous membranes which often poses a major therapeutic challenge due to its refractory course. Novel pathogenesis-based therapies are urgently needed. As several studies have shown that IL-17 may contribute to LP pathogenesis, we investigated whether therapeutic targeting of IL-17 + T cells leads to clinical improvement of mucosal and cutaneous LP lesions. A total of five patients with lichen planus w… Show more

“…Since Mrp8 and Mrp14 are detectable in kidney and in cutaneous lesions of patients with lupus erythematosus as well as in skin lesions of patients with lichen planus and psoriasis, it is conceivable that these proteins play a role in the induction of Tc17 cells in these disorders [72]. Consistently, an involvement of Tc17 cells has been recently shown also for lichen planus, an inflammatory skin and mucous membranes disease, which could be ameliorated by targeting of the IL-23-IL-17 axis [73], similarly to clinical success in the treatment of psoriasis [74].…”

“…The correlation with the other inflammatory cytokines did not reach statistical significance. The important role of IL‐17 in oral mucosal inflammation is supported by evidence of favourable response to the therapeutic targeting of IL‐17 + T cells in recalcitrant erosive lichen planus (Solimani et al, 2019).…”

“…Nowadays, patients with this chronic inflammatory disease are frequently treated with biologicals, either of the first generation (anti-TNF) or the second generation (anti-IL-17/anti-IL-23) (47). In recent years, our better understanding of further inflammatory skin diseases such as lupus erythematosus (LE), lichen planus (LP) or atopic dermatitis (AD) also resulted in the development of novel treatment strategies (811). At the same time, we have gained more insight into the role of proteins propagating the intracellular effects of activated cytokine receptors (12).…”
