Therapeutic Ultrasound-Enhanced Immune Checkpoint Inhibitor Therapy

Abstract: Immune checkpoint inhibitors (ICIs) are designed to reinvigorate antitumor immune responses by interrupting inhibitory signaling pathways and promote the immune-mediated elimination of malignant cells. Although ICI therapy has transformed the landscape of cancer treatment, only a subset of patients achieve a complete response. Focused ultrasound (FUS) is a noninvasive, nonionizing, deep penetrating focal therapy that has great potential to improve the efficacy of ICIs in solid tumors. Five FUS modalities have … Show more

“…Despite ultrasound being known to be a noisy imaging modality compared to various Ultrasound-Based AI for COVID-19 Detection other imaging modalities with exceptional image quality (i.e., CT, magnetic resonance imaging (MRI), X-ray, etc.) (Park et al, 2020), it stands out for being a low-cost, mobile, and, above all, non-ionizing medical imaging technology (Yuan et al, 2021). Because ultrasound is radiation-safe, it is the preferred imaging modality for children and pregnant women (Prentice et al, 2022) and has been widely used in the detection and severity assessment of COVID-19 for the same patient group (Allinovi et al, 2020).…”

“…explicitly mentioned. In non-neurological conditions, FUS+MB exposure has been shown to stimulate immune responses that may be beneficial (e.g., in metastatic cancer) or directly induce anti-inflammatory effects at the target site [82][83][84][85]. Recent studies have also shown that immunomodulation can be successfully used to treat GBM, and that FUS+MBs can be effective in inducing targeted immune effects and in delivering immunotherapeutics with promising results [86,87].…”

“…While there has been a significant and deserved emphasis on focused ultrasound to transiently permeabilize the blood–brain barrier, ablative ultrasound therapies can also enhance immune checkpoint inhibition [ 188 ]. Thermal ablative ultrasound therapy uses high-intensity focused ultrasound to increase the local temperature to 60 °C or higher to induce coagulative necrosis.…”
