Therapy of HIV Infection: Current Approaches and Prospects

Abstract: The human immunodeficiency virus type 1 (HIV-1) is the causative agent of one of the most dangerous human diseases – the acquired immune deficiency syndrome (AIDS). Over the past 30 years since the discovery of HIV-1, a number of antiviral drugs have been developed to suppress various stages of the HIV-1 life cycle. This approach has enables the suppression of virus replication in the body, which significantly prolongs the life of HIV patients. The main downside of the method is the development of viral resist… Show more

“…Antiretroviral therapy (ART) is used to suppress HIV infection. It comprises pharmacological drug classes including nucleoside reverse transcriptase inhibitors (NRTIs), nonnucleoside reverse transcriptase inhibitors (NNRTIs), protease inhibitors (PIs), integrase strand transfer inhibitors (INSTIs), fusion inhibitors, and chemokine receptor antagonists [10,11]. The WHO 2013 ART guidelines recommended a fixed-dose, single-tablet regimen containing two NRTIs [tenofovir disoproxil fumarate (TDF) + lamivudine (3TC)] plus one NNRTI [efavirenz (EFV)] (TLE600 STR) as the preferred first-line regimen for ART-na€ ıve adults and adolescents in low-and middle-income countries (LMICs) such as India [12].…”

“…Acquired immunode ciency syndrome (AIDS), caused by the human immunode ciency virus (HIV), is one of the most serious infectious diseases in the world [1], with no cure or effective vaccine so far. Highly active antiretroviral therapy (ART), developed in 1996, is still the only effective method to inhibit HIV replication.…”

“…Drugs that directly inhibit virus replication have become mainstays in the treatment of chronic viral infections such as HIV/AIDS, 1 hepatitis C virus (HCV), 2 hepatitis B virus, herpesvirus and acute infections such as influenza. RNA viruses such as HIV-1 and HCV possess ideal targets for virus inhibition such as RNA-dependent RNA polymerase and protease.…”
