Thermoresponsive Hydrogel-Based Local Delivery of Simvastatin for the Treatment of Periodontitis

Abstract: Except for routine scaling and root planing, there are few effective nonsurgical therapeutic interventions for periodontitis and associated alveolar bone loss. Simvastatin (SIM), one of the 3-hydroxy-3-methylglutaryl-cosenzyme A reductase inhibitors, which is known for its capacity as a lipid-lowering medication, has been proven to be an effective anti-inflammatory and bone anabolic agent that has shown promising benefits in mitigating periodontal bone loss. The local delivery of SIM into the periodontal pocke… Show more

“…Drug release can be changed by modulating various composition parameters, such as polymer materials, polymer concentration and sustained-release agents [ 24 , 25 ]. Correspondingly, the varying gel degradation rate can be used to modulate the drug release rate [ 26 ]. The MC was added as a sustained-release agent to gel.…”

“…Current treatments for periodontitis include non-surgical and surgical debridement, coupled with local or systemic delivery of antibiotics. 2 While these treatments remove the source of infection (dental plaque), they don't address the subsequent immune-inflammatory healing response. Cell transplantation has been applied to attenuate periodontitis in animal models.…”

“…Compared with conventional wound dressings, hydrogels are considered to be wound treatment materials with great potential due to their good biocompatibility, moist environment, absorption of exudate and predictable degradation rate to relieve the pain of patients. [7][8][9] In particular, thermo-sensitive hydrogels are one of the most popular smart hydrogels due to their rapid and reversible temperature-dependent sol-gel transition behavior. 9 Thermosensitive hydrogels can be designed to load bio-active factors or cells by simple mixing of solutions at low temperature and form a hydrogel at body temperature as a continuous drug release carrier, 10,11 with this process allowing bio-active factors or cells to easily enter the body in a minimally invasive manner.…”
