Three-Dimensional Human Neural Stem Cell Models to Mimic Heparan Sulfate Proteoglycans and the Neural Niche

Abstract: Heparan sulfate proteoglycans (HSPGs) are a diverse family of polysaccharides, consisting of a core protein with glycosaminoglycan (GAG) side chains attached. The heterogeneous GAG side-chain carbohydrates consist of repeating disaccharides, with each side chain possessing a specific sulfation pattern. It is the variable sulfation pattern that allows HSPGs to interact with numerous ligands including growth factors, cytokines, chemokines, morphogens, extracellular matrix (ECM) glycoproteins, collagens, enzymes,… Show more

“…They also act as membrane-bound receptors. These molecules include heparan sulfate proteoglycans (HSPGs), comprising two distinct families: syndecans (4 members) and glypicans (6 members) [ 99 , 100 ]. Numerous articles give detailed descriptions of the structure and functioning of HSPGs [ 99 , 101 – 106 ].…”

“…Glypicans are globular glycosylphophatidylinositol (GPI) anchored proteins [ 99 , 111 ]. Typically, glypicans have an N-terminal cysteine-rich domain similar to that of Frizzled receptors and mediate Wnt binding [ 101 ].…”

“…Peall et al thus present an overview on the current knowledge in three-dimensional human neural stem cell models, discussing their application in exploring HSPGs and the neural extracellular niche. 8 Better understanding of the role of HSPGs in neurologic pathologies could advance therapeutic approaches. Moreover, these models might be applicable to HS research on other stem cell types and their extracellular niche.…”
