Thrombospondin-1 mediates distal tubule hypertrophy induced by glycated albumin

Abstract: Diabetic nephropathy is characterized by early hypertrophy in both glomerular and tubuloepithelial elements. However, no studies to date have established a direct causal link between hyperglycaemia and renal hypertrophy. Our previous studies have found that high glucose does not induce cellular hypertrophy or expression of TGF-beta1 (transforming growth factor-beta1) in distal renal tubule cells [Yang, Guh, Yang, Lai, Tsai, Hung, Chang and Chuang (1998) J. Am. Soc. Nephrol. 9, 182-193]. In the present study, w… Show more

“…48 TSP1 expression is stimulated by glycated albumin treatment of Madin-Darby canine kidney renal tubular cells in vitro, and treatment of cells with anti-TSP1 antibodies reduced cellular hypertrophy and TGF-␤ activity. 38 TSP1 mRNA expression is also increased in aged and obese Zucker rat tubulointerstitium. 56 In streptozotocin-treated endothelial nitric oxide synthase knockout mice, which develop diabetic nephropathy in addition to hypertension, TSP1 expression is increased and correlates with TGF-␤ expression and tubulointerstitial injury.…”

“…64 Treatment of mesangial cells with high levels of amino acids increases TSP1 expression and TSP1-dependent TGF-␤ activity, 65 and glycated albumin increases TSP1 expressed by cultured renal tubular cells, with increases in TSP1-dependent TGF-␤ activation. 38 These data suggest that TSP1 has a significant effect on proteinuria through regulation of TGF-␤ activity.…”

“…17 In addition, the promoter of thrombospondin-1 and -2, the first naturally occurring angiostatic agents discovered, contain NF-kB binding sites. 18 The role of NF-kB in the progression of cell cycle has also been investigated. NF-kB is able to induce expression of both activators and inhibitors of cell cycle progression.…”
