Thymoquinone (TQ) regulates cyclooxygenase-2 expression and prostaglandin E2 production through PI3kinase (PI3K)/p38 kinase pathway in human breast cancer cell line, MDA-MB-231

Abstract: Thymoquinone (TQ), a drug extracted from the black seeds of Nigella sativa, has been shown to exhibit antiinflammatory, anti-oxidant, and anti-neoplastic effects in numerous cancer cells. The effects of TQ on cyclooxygenase-2 (COX-2) expression and prostaglandin E 2 (PGE 2 ) production in MDA-MB-231, however, remain poorly understood. Western blot analysis and immunofluorescence staining were performed to study the expression levels of inflammation regulatory proteins in MDA-MB-231. PGE 2 assay was conducted t… Show more

“…Specifically, TQ in lower concentrations induced phosphorylation of Akt while higher concentrations inhibited p-Akt. TQ stimulatory effects on p-Akt were previously observed in MDA-MB-231 breast cancer cells ( Yu and Kim, 2012 ) while the inhibition of p-Akt by TQ was previously confirmed in various cancer models ( Koka et al, 2008 ; Yi et al, 2008 ; Arafa et al, 2011 ; Hussain et al, 2011 ; Rajput et al, 2013 ; Khan et al, 2019 ). The observed TQ hormetic effects where low concentration increased levels of p-NF-κB p65 in LAMA-84r cells and p-Akt in both LAMA-84 cell lines were probably due to compensatory mechanisms activated by cells.…”

“…The overexpression of COX2 plays a crucial role as prostaglandin up-regulates angiogenesis and increases the resistance to apoptosis (Costa et al, 2002 ; Santos and Costa-Pereira, 2011 ). COX2 is reported for the inhibition of some clinical behavior of tumors (Banerjee et al, 2009 ; Yu and Kim, 2012 ). The inhibitor of COX2 shows a significant effect on its inhibition, while it also possess some side effect on other tissues, therefore in recent years natural product received attention for its choice to prevent cancer through the COX2 inhibitory mechanism.…”

“…The overexpression of COX2 plays a vital role in the upregulation of angiogenesis by prostaglandin and also shows effect in apoptosis by increasing the resistance to apoptosis [ 91 ]. COX2 also shows the inhibition of clinical behavior of some tumors [ 92 ]. However, COX2 inhibition is a critical step in the prevention of cancer.…”
