Thyroid Pathology inPTEN-Hamartoma Tumor Syndrome: Characteristic Findings of a Distinct Entity

Abstract: There were no morphologic differences between the thyroid findings in CS and BRRS. Also, there was no correlation between specific PTEN germline mutations (exons 5, 6, and 8) and pathologic findings. Distinctive and characteristic findings in PHTS include multiple unique adenomatous nodules in a background of LT, and C-cell hyperplasia; it is vital that pathologists recognize the classical histologic features of this syndrome to alert clinicians to the possibility of this syndrome in their patients.

“…The progression of skin hamartoma in Pten D mice was evaluated according to the pathological criteria of PHTS as described in previous studies (Supplemental Fig. 1B; Nelen et al 1996;Eng 2000;Laury et al 2011). The median onset of disease in Pten D mice was 62 d (Fig.…”

“…The progression of mucocutaneous lesions in mice was monitored according to the operational diagnosis criteria of PHTS (Nelen et al 1996;Eng 2000;Laury et al 2011). The facial papillomatous, acral keratoses, and palmoplanta keratoses (limbs and paws) were each graded from 0 (normal) to 4 (severe), and a summed score was calculated accordingly.…”

“…Thyroid pathologic findings in this syndrome typically affect follicular cells. 50,51 Follicular carcinoma, with a frequency of 5-10%, is a major diagnostic criterion for the diagnosis of CS; multinodular goiter (nodular hyperplasia), multiple adenomatous nodules, and follicular adenomas are minor criteria, with a frequency of 50-67%.…”

“…The majority of thyroid lesions occurring in PHTS are follicular carcinoma; PTC is rarely been associated with this entity. 50,53 The occurrence of papillary microcarcinoma in CS is usually considered to be a chance occurrence as such tumors may be present in up to 30% of the adult population. 50 MTC is not considered as a part of the spectrum of PHTS; however, previous studies, including studies conduct by us 50,53 and others, 51 have identified CCH in individuals affected with this syndrome.…”
