“…The next year, apparently unaware of this work, Benington and Morin performed the same synthesis, and shortly thereafter a note was published recapitulating the seemingly obvious benzyl alcohol–chloride–nitrile–amine route that had been discarded as inefficient in the 1930s, now using LAH reduction, but less satisfactorily . This methodology was used by Dornow and Petsch, with K[ 14 C]CN, to prepare radioactive mescaline for an early pharmacokinetic study (Scheme F) . LAH was also employed to obtain mescaline by reduction of 3,4,5-trimethoxyacetamide, prepared in turn from the 3,4,5-trimethoxybenzoyl chloride via the diazoketone.…”

“…39 This methodology was used by Dornow and Petsch, 40 with K[ 14 C]CN, to prepare radioactive mescaline for an early pharmacokinetic study (Scheme 1F). 41 LAH was also employed to obtain mescaline by reduction of 3,4,5-trimethoxyacetamide, prepared in turn from the 3,4,5-trimethoxybenzoyl chloride via the diazoketone. This method, in spite of its relatively poor overall yield, was used to prepare N-methylmescaline and N,Ndimethylmescaline (trichocereine) (Scheme 1G).…”

“…The question arises whether LSD may have an indirect action which would explain the production of psychic effects. Block (2,3) has reported that the mental effects of mescaline in humans occur at about thirty minutes after its administration, i.e., at a time when in animals no more traces of this chemical could be detected in the brain. On the basis of extensive biochemical experiments in vitro and in animals, he showed that mescaline was converted, in the liver, into a new, protein-bound compound; a reaction which required about thirty minutes for its completion.…”
