Total Flavonoids of Rhizoma Drynariae Ameliorate Bone Growth in Experimentally Induced Tibial Dyschondroplasia in Chickens via Regulation of OPG/RANKL Axis

Abstract: Background:Rhizoma Drynariae, traditional Chinese herb, is widely used to treat and prevent bone disorders. However, experimental evidence on the use of Rhizoma Drynariae extract, total flavonoids of Rhizoma Drynariae (TFRD) to treat tibial dyschondroplasia (TD) in chickens and its underlying mechanisms have not been investigated.Purpose: To evaluate the therapeutic effect of TFRD on leg disease caused by TD and elucidate its mechanisms in modulating the bone status. Methods: Thiram-induced chicken TD model has… Show more

“…Our results indicated that tibial bone strength, Ca and P metabolism disorder, and bone formation marker ALP decreased in TD broilers. The results of this study are consistent with the findings of Xu et al [ 37 ] (2018) and Liu et al [ 42 ] (2021). Many studies have proved that TCM has the function of tonifying the kidneys and strengthening the bones.…”

“…TD is a susceptible disease of the tibial tarsus affecting the proximal growth plate of the tibia in fast-growing poultry, and its prevention and treatment have not been well defined. Numerous studies have confirmed that traditional Chinese herbal medicine plays an important role in improving bone development, such as total flavonoids of Rhizome drynariae [ 37 ], Puerarin [ 38 , 39 ], and Astragalus membranaceus [ 18 ]. We investigated the therapeutic effects of MOP on TD broilers, and the results showed that MOP could improve tibia-related indicators through BMP/Smads signaling pathway, improve the disturbance of Ca and P metabolism caused by TD, and increase the antioxidant capacity of the chickens.…”

“…AFB1 is a class of highly toxic mycotoxins, and the waste of feed ingredients and decreased animal performance caused by AFB1 contamination have brought heavy economic losses to the livestock industry [ 36 ]. As a flavonoid herbal medicine extract, TFRD has various bioactive effects, including immunomodulatory, anti-inflammatory, antioxidant, and anti-apoptotic effects [ 1 , 5 , 29 , 31 ]. In this present experiment, the main active ingredient of TFRD is Rutin .…”

“…TFRD (#K20798) had been acquired from Xi’an Kailai Biological Engineering Co., Ltd. (Xi’an, China). The compositional analysis of TFRD is shown in Figure S1 and Table S1 , and its main component was detected by liquid chromatography-mass spectrometry (LC-MS) analysis as Rutin (97.82%) [ 29 ]. Hematoxylin and eosin (HE) and Periodic Acid-Schiff (PAS) were purchased from Servicebio ® (Wuhan, China).…”

“…TFRD extract is derived from the roots of Drynaria roosii Nakaike 74 . It has been indicated to be useful in the treatment and prevention of bone disorders by increasing the proliferation and differentiation of osteoblasts and promoting bone strength 75 , 76 . In the present study, after 14 days of TFRD treatment, TD symptoms in the TFRD group were significantly reduced, accompanied by the restoration of GM homoeostasis, and the GLU levels and the pancreas almost returned to normal.…”
