(1) Background: Although Platycodon grandiflorum saponins exhibit many beneficial biological effects in various diseases and conditions, how they protect nerve cells against neurodegenerative diseases and Alzheimer’s disease (AD) pathology is unknown. We investigated whether P. grandiflorum crude saponin (PGS) protects neurons from neurodegeneration caused by amyloid beta (Aβ)-induced oxidative stress. (2) Methods: Hippocampal neuron HT-22 cells were used in the in vitro experiment, and AD mice (5XFAD mice) were used as the in vivo model. Intracellular reactive oxygen species (ROS) was stained with DCF-DA and assessed using fluorescence microscopy. To elucidate the mechanism underlying neuroprotection, intracellular protein levels were assessed by western blotting. In 5XFAD mice, an animal model of AD, nerve damage recovery due to the induction of Aβ toxicity was evaluated by histological analysis. (3) Results: PGS attenuates Aβ-induced neurotoxicity by inhibiting Aβ-induced reactive oxygen species (ROS) production and apoptosis in HT-22 cells. Furthermore, PGS upregulated Nrf2-mediated antioxidant signaling and downregulated NF-κB-mediated inflammatory signaling. Additionally, PGS inhibited apoptosis by regulating the expression of apoptosis-associated proteins. In addition, PGS ameliorated Aβ-mediated pathologies, leading to AD-associated cognitive decline. (4) Conclusions: Taken together, these findings suggest that PGS inhibits Aβ accumulation in the subiculum and cerebral cortex and attenuates Aβ toxicity-induced nerve damage in vitro and in vivo. Therefore, PGS is a resource for developing AD therapeutics.

In this study, general Sibjeondaebohwan (G-SDH) and high-temperature aged Sibjeondaebohwan (HTA-SDH) samples were prepared using ten herbal medicinal plants including balloon flower. The nutritional components (five nutrients, fatty acids, and free amino acids), phytochemicals (phenolic acids and flavonols), radical scavenging activities (DPPH, ABTS, and hydroxyl), digestive enzyme inhibitory activities (α-glucosidase and pancreatic lipase), and immune system activation of G-SDH and HTA-SDH were analyzed and compared. The contents of crude ash, fat, and protein, carbohydrate, total fatty acid, and total free amino acid of G-SDH were significantly higher than those of HTA-SDH. The major free amino acids and fatty acids in G-SDH were aspartic acid and oleic acid and those in HTA-SDH were glutamic acid and oleic acid. Compared to G-SDH, higher concentrations of total polyphenols, total flavonoids, Mailliard reaction products, total phenolic acids and total flavonols of HTA-SDH were estimated. After high-temperature aging, the total phenolic and flavonol contents increased from 272.03 to 303.31 μg/g and 310.71 to 372.58 μg/g, respectively. The DPPH, ABTS, and hydroxyl radical scavenging activities and the α-glucosidase, α-amylase, and pancreatic lipase inhibition activities in HTA-SDH were 85.65%, 89.93%, and 63.69%, and 61.06%, 34.21%, and 64.89%, respectively, which were considerably higher than those in the G-SDH. In addition, HTA-SDH showed the effect of controlling immune function.

Antioxidant and anticancer activities of Adenophora triphylla leaf and root extracts

The root of Adenophora triphylla is a highly valued medicinal resource that is used to prevent human obesity, cancer, and inflammation, whereas young leaves or sprouts of A. triphylla are used as food ingredients. In this study, we compared the antioxidant and anticancer activities of 70% ethanol extracts of A. triphylla roots and leaves. The leaf extract exhibited stronger 2,2-diphenyl-1-picrylhydrazyl (DPPH)-radical scavenging activity, reducing power, and oxygen radical absorbance capacity (ORAC) than the root extract. Furthermore, the leaf extract was observed to be a potent source of anticancer compounds that were effective against A549 (lung cancer), LNcaP (prostate cancer), SKOV3 (ovarian cancer), and Caco-2 (colorectal cancer) cells. These results indicate that not only the roots but also the leaves of A. triphylla can serve as valuable sources of functional materials in the pharmaceutical industry.
