RESUMO: "Avaliação das atividades moluscicida e cercaricida (Schistosoma mansoni) dos extratos de Croton floribundus e do ácido caurenóico". Extratos de Croton fluribundus (Euphorbiaceae), ácido caurenóico e dois derivados do ácido caurenóico foram avaliados como moluscicida, cercaricida e também foi verificada a letalidade destas amostras frente a larvas de Artemia salina Leach. Nestes ensaios foram observadas significantes atividades moluscicida e cercaricida associadas a uma reduzida toxicidade frente ao camarão de água salgada. Unitermos: Biomphalaria glabrata, Croton floribundus, Artemia salina, ácido caurenóico, Schistosoma mansoni, cercárias.ABSTRACT: Lethality of the extracts of Croton floribundus (Euphorbiaceae), a medicinal plant from south Brazil, and of the kaurenoic acid, an isolated compound, and two of its derivatives against adult Biomphalaria glabrata snails, Schistosoma mansoni cercariae and Artemia salina Leach. brine shrimp larvae are reported. Both extracts and the isolated compound showed significant molluscicidal and cercaricidal activities and reduced toxicity in brine shrimp assays.

ABSTRACT. The search for new and effective antitumor agents with fewer cytotoxic side effects on normal tissue has increasingly become important. Lapachol, a natural organic compound isolated from the lapacho tree (Tabebuia avellandedae), is chemically identified as belonging to the naphthoquinone group and is known for its antiinflammatory, analgesic and antibiotic properties, although there are questions about its effectiveness for treating neoplasic cells. We evaluated the antitumoral effects of lapachol by testing for clones of epithelial tumors in Drosophila melanogaster. Seventy-two-hour old larvae bred from wts/TM3, Sb 1 females and mwh/mwh males, were treated with different concentrations of lapachol (20, 40 and 60 µg/mL). Lapachol alone did not significantly increase the number of epithelial tumors. However, lapachol did significantly reduce the number of tumors provoked by doxorubicin.

Background: Continuous attempts are being made to develop new and more effective drugs for treatment of schistosomiasis. Curcuma longa is a medicinal plant that contains yellow pigments known as curcuminoids (curcumin, bidesmetoxi-curcumin and monodesmetoxi-curcumin), with curcumin as the highest component. The main biological effect depends on curcumin content. Curcumin has low bioavailability and poor-water solubility. Preparation of C. longa (Cl) nanoemulsion was attempted to enhance solubility and bioavailability of curcumin. Objective: The production of Cl nanoemulsion, and study of its pharmacokinetic properties (namely solubility and intestinal bioavailability) and anti-helminthic effects on Schistosoma mansoni cercariae, schistosomules and adults. Methodology: Cl nanoemulsion was prepared from ethanol extract of crude powder of Cl rhizome. Characterization was done by transmission electron microscope (TEM), testing solubility and intestinal bioavailability. Schistosomicidal effects were performed by in vitro assay. Results: Cl nanoemulsion showed increased solubility and bioavailability compared to ethanol extract of the tested plant; as well as time and dose dependent schistosomicidal effects on cercariae and 24h-old schistosomules of S. mansoni. In addition, it exhibited an optimal activity against the adult stage with decreased motor activity of the worms. Conclusion: This study proves that Cl nanoemulsion has enhanced solubility and bioavailability, as well as schistosomicidal activity.
