Toxicity of silver nanoparticle in rat ear and BALB/c 3T3 cell line

Abstract: Background: Silver nanoparticles (AgNPs) displayed strong activities in anti-bacterial, anti-viral, and anti-fungal studies and was reportedly efficient in treating otitis media .The potential impact of AgNPs on the inner ear was missing.

“…Regardless of the outcome of the in vitro study, we should also remember that the mouse 3T3 cell culture line may be many times more sensitive than the cells examined under in vivo conditions . Another important recommendation when working with the 3T3 cells is that the response of 3T3 fibroblasts to oxidative stress, which is one of the mechanisms of cell death, may undergo alterations after long‐term cell exposure to endocrine disruptors …”

“…The utility of the 3T3 cell line has been shown in many other biological and toxicological studies, using various experimental approaches. For example, 3T3 fibroblasts have been selected for the following purposes: 1/investigating the activity of bacterial toxins, antioxidants and cytokines, 2/evaluating the toxicity of drugs, e‐waste leachate, nanomaterials and nanoparticles or 3/studying the beneficial effects of electromagnetic radiation in living cells . These cells can also serve as a tool for the verification and validation of techniques assessing mitochondrial toxicity .…”

“…Neutral red uptake assay, originally developed by Borenfreund and Puerner , was performed using the standard protocol of OECD Guidance document 129 and as described in Zou et al . . Briefly, 25 μg/ml NR solution was added to the cells and blanks and the plates were incubated at 37°C, 5% CO 2 for 3.0 ± 0.1 hr.…”

“…In in vitro studies, Ag NPs were used for MRI imaging. Their toxic effect on the structure of the ear was investigated in rats; they reduced hearing frequency in rats, were effective on the mitochondrial membrane and increased ROS level [175].…”

“…After 5 h, NP penetration into cells was observed which caused mitochondrial dysfunction; the toxicity was dose-and time-dependent [175].…”
