Transcriptional Activation of Human CDCA8 Gene Regulated by Transcription Factor NF-Y in Embryonic Stem Cells and Cancer Cells

Abstract: Background:The regulation of CDCA8 gene expression remains unclear. Results: CDCA8 promoter is activated in hESCs and cancer cells, in which NF-Y is a positive-regulator by binding to the CCAAT-box. Conclusion: CDCA8 is transcriptionally activated in hESCs and cancer cells and is positively regulated by NF-Y. Significance: Characterization of CDCA8 regulation provides a better understanding of its biological functions.

“…We focused on another cell cycle regulator, CDCA8, after the screening of CHIP–CHIP assays (data not shown). Several studies showed CDCA8 was involved in tumorigenesis, however, the exact function of CDCA8 in cancer development is unknown (Bi et al, ; Dai et al, ; Jiao et al, ).…”

“…Western blot analyses were carried out as previously described (31). After collecting total protein with RIPA buffer (Pierce, Rockford, IL, USA), total protein was separated on SDSpolyacrylamide gels, then transferred to PVDF membrane (EMD Millipore, Billerica, MA, USA).…”

“…22 Subunit NF-YA of transcription factor NF-Y (nuclear factor Y), which, as it will be discussed in the following section, up-regulates proteasome sub-units, is involved in positive regulation of cycle regulator CDCA8 in embryonic cells, and thus, a program of proteasome up-regulation, satisfying increased metabolic needs, may be coordinately regulated with the cell cycle. 48 In addition, a short isoform of NF-YA regulates the pluripotency program itself in embryonic cells. 49 This prerequisite of high proteasome activity for gene priming is not shared by normal adult tissue stem cells and CSCs which retain a more restricted ability to differentiate to different cells of tissue of origin or to different bulk cancer cells (Figure 3).…”
