Transcriptional mechanism of vascular endothelial growth factor-induced expression of protein kinase CβII in chronic lymphocytic leukaemia cells

Abstract: A key feature of chronic lymphocytic leukaemia (CLL) cells is overexpressed protein kinase CβII (PKCβII), an S/T kinase important in the pathogenesisIn addition to CLL, overexpression of PKCβ II is also observed in B-lymphocyte malignancies such as diffuse large B cell and mantle cell lymphoma 12,13 , and in epithelial tumours such as carcinoma of the colon 14 and breast 15 . Indeed, like CLL, the development of colon cancer is intrinsically linked with overexpressed PKCβ II 16,17 . Therefore, understanding ho… Show more

“…Our data show that elevated PKCβ expression in PKCα-KR-transduced cells is driven by elevated Sp1 binding activity at the prkcb promoter. This is similar to what is observed in human CLL cells [ 17 ] and suggests that the mechanisms controlling PKCβ expression are accurately modelled by our system. A further similarity is demonstrated by our experiments using mtm to target the DNA binding activity of SP1.…”

“…The transcription factor SP1 plays a central role in PRKCB gene regulation in human CLL [ 17 ]. We were interested in determining whether the prkcb gene was similarly regulated by Sp1 in our CLL mouse model.…”

“…19 The analysis was conducted as previously described. 47 The Jetset database 48 was employed to select more reliable probes from the Affymetrix HG-U133 Plus 2.0 chip for the common genes found. Only expression data from PBMC samples ( n = 1120) and neutrophils ( n = 228) were selected.…”
