Transcriptional variation in glucosinolate biosynthetic genes and inducible responses to aphid herbivory on field-grownArabidopsis thaliana

Abstract: 25Recently, increasing attempts have been made to understand how plant genes function in 26 natura studies. To determine whether plant defense genes are activated under multiple biotic 27 stimuli, we combined a high-throughput RNA-Seq with insect survey data on 19 accessions 28 of Arabidopsis thaliana growing on the field site of Switzerland. We found that genes with 29 GO annotations "glucosinolate biosynthetic process" and "response to insects" were the most 30 significantly enriched, exhibiting largely vari… Show more

“…This enzymatic activity transfers a methyl group to a wide range of substrates, including several low molecular weight metabolites acting as plant hormones, and is key in phenylpropanoid, flavonoid and glucosinolate plant metabolic pathways 38 , 39 . Another notable expansion was found in orthogroup OG0007231, which contains seven genes in Moringa, five in Arabidopsis and none in the rest of species, encoding for members of the large family of GDSL-type esterases/lipases, formed by hydrolytic enzymes with multifunctional properties such as broad substrate specificity 40 , including Arabidopsis epithiospecifier modifier 1 (ESM1), which represses nitrile formation and favors isothiocyanate production during glucosinolate hydrolysis 41 , 42 .…”

“…In contrast, no AOP3 loss-of-function mutant has been reported to date, which might be linked to specific defensive properties of AOP3-modified GSL present in the seeds and thus the seedlings of all A. thaliana genotypes. Constitutive expression of AOP3 in leaves does not occur in all accessions but can be induced upon attack 50 .…”

“…To further identify their biological functions, these DEGs were subjected to GO enrichment analysis. GO is a model biological framework that defines concepts used to describe gene function, and relationships between these concepts (Ana et al 2005;Sato et al 2019;Wang et al 2019). Based on sequence homology, DEGs were assigned to three general categories: cellular component (CC), molecular function (MF), and biological process (BP).…”
