Transcutaneous immunization with an outer membrane protein of Porphyromonas gingivalis without adjuvant elicits marked antibody responses

Abstract: Since 40k-OMP-specific IgG inhibited the coaggregation of P. gingivalis with Streptococcus gordonii, and the hemagglutinin activity of P. gingivalis, TCI with the 40k-OMP may be important as an adjuvant-free immunogen for the prevention of chronic periodontitis.

“…Previous studies showed that a monoclonal antibody (MAb) to 40-kDa OMP inhibits coaggregation by P. gingivalis and promotes complement-mediated bactericidal and opsonic activity for the phagocytosis of P. gingivalis (1,21,46). Furthermore, rats that are transcutaneously immunized with a 40-kDa OMP vaccine have significantly diminished P. gingivalis-induced abscess formation (24). Therefore, the development of a mucosal 40-kDa OMP vaccine for humans may be a significant milestone in the search for an effective vaccine against P. gingivalis infection.…”

“…(13,14) Furthermore, nasal or transcutaneous immunization with 40k-OMP prevented alveolar bone loss or abdominal abscess formation in response to a P. gingivalis challenge. (15,16) Therefore, the use of 40k-OMP as a mucosal vaccine antigen may be beneficial in the development of a periodontal vaccine and for the preventive therapy of cardiovascular disease (CVD).…”

“…Second, transcutaneous or nasal immunization of mice with 40-kDa OMP and CT has shown to induce serum IgG, IgA and IgG in saliva [82,83]. Presently, adjuvant-free transcutaneous immunization of rats with 40-kDa OMP induced a similar immune response and protection against subcutaneous challenge with P. gingivalis 381, a nonvirulent strain [84]. To circumvent the adverse effects of the CT, a nontoxic chimeric variant of CT could be used.…”
