“…Many of these studies have been performed on cell lines where the resulting altered cells (transformed cells) could be cloned and subcultured for further analysis. Except for hamsterembryo cells, which have been studied extensively (Berwald & Sachs, 1965;DiPaolo & Donovan, 1967;DiPaolo et al, 1971;Huberman et al, 1972), little is known about the effect of polycyclic aromatic hydrocarbons on primary or secondary cultures of other animal cells.…”

“…Whereas, as a number of authors have shown [20][21][22][23] various metabolites of polycycling carcinogenic hydrocarbons, resulting from the activity of certain enzymes upon primary hydrocarbons are responsible for the trans forming and toxic effects of the above agents [23]. More, L iberman and D ip p l e [24] have shown in direct experiments that human peripheral blood lymphocytes contain enzymes catalizing the conversion of 7-brommethylbenz-(a)-anthracene into a compound which forms a covalent bond with the lymphocyte DNA.…”

“…For the quantitative transformation assay, 300 secondary hamster cells are placed on an irradiated hamster feeder layer (DiPaolo, et al, 1971a(DiPaolo, et al, , 1971b. After the cells have attached to the plastic Petri dish surface, they are exposed to either one treatment or a sequence of treatments.…”

“…Cells may be protected from the cytotoxic effect of polycyclic hydrocarbon carcinogens by the inhibition of the induction of arylhydrocarbon hydroxylase (AHH) (Alfred and DiPaolo, 1968;Diamond and Gelboin, 1969;Gelboin et al, 1970). Protection by benz(a)anthracene (BA) and inhibition of 7,12-dimethylbenz(a)anthracene (DMBA) initiated cell lysis, cell division, and DNA synthesis were first reported in 1968 (Alfred and DiPaolo, 1968) and elaborated upon in 1971(DiPaolo et al, 1971c. These chemicals give different results, depending upon the carcinogen, the cells used, and the protocol, particularly in terms of the time of addition of the various chemical agents.…”
