Transmission of plasmid-borne and chromosomal blaCTX-M-64 among Escherichia coli and Salmonella isolates from food-producing animals via ISEcp1-mediated transposition

Abstract: Objectives To clarify the transmission mechanism of the blaCTX-M-64 gene between Escherichia coli and Salmonella isolates from food animals. Methods A total of 329 E. coli and 60 Salmonella isolates collected from food animals in 2016 were screened for the presence of blaCTX-M-64 genes. The blaCTX-M-64-positive isolates were typed and plasmid and chromosome DNA was sequenced to determine the genetic context of blaCTX-M-64 and… Show more

“…The contigs carrying bla CTX-M-14 in the four chicken isolates showed high similarity to each other and to the backbone of the IncHI2 plasmid, while beef isolate 19M harbored a chromosomal bla CTX-M-14 (Figure 1C). This highlights the potential role of ISEcp1 in mobilization of bla CTX-M-14 regardless of whether the gene was located on plasmids or the chromosome (Zhao et al, 2020). Both variants of fosA, fosA3 in beef isolate 21M and fosA4 in chicken isolate 82CH, were located downstream to IS26 belonging to the IS6 family (Figures 1D, E), which might be involved in their mobilization.…”

“…IS Ecp1 appeared to be able to use IRL in combination with a sequence beyond its IRR end to move an adjacent region, yielding the transfer of the entire transposon unit [ 59 , 60 ]. IS Ecp1 -mediated transposition of bla CTX-M-64 , bla CTX-M-2 , bla CTX-M-3 , and bla TEM-1b have been demonstrated [ 61 , 62 , 63 ]. Moreover, IS Ecp1 could capture DNA regions with different sizes and simultaneously transfer adjacent regions [ 59 ].…”

“…The pap2 gene was always present downstream of mcr-1 except for QH20-4D2-1 contig_81 (JAJDKI010000081), which contained bla CTX-M-64 , an independent resistance region of the ESBL resistance gene. This region was located in ISEcp1-bla CTX-M-64 -Δorf477, a typical transposon unit, of which the ISEcp1 insertion sequence can mediate the transposition of many types of the bla CTX-M gene (Zhao et al, 2020). Except in the case of QH20-4B1-2 contig_139 (JAJDKH010000139), the resistance gene mcr-1 was integrated at a location downstream to the nikB gene, which is identical to that of the majority of mcr-1-positive IncI2-type plasmids reported previously.…”
