Transport Dynamics of MtrD: an RND Multidrug Efflux Pump fromNeisseria gonorrhoeae

Abstract: Antibiotic-resistant gonorrheal infections are an urgent health concern. The MtrCDE system confers multidrug resistance to Neisseria gonorrhoeae, an obligate human pathogen, and the causative agent of the sexually-transmitted infection gonorrhea. The inner membrane pump MtrD effluxes a variety of hydrophobic and amphiphilic substrates and thereby confers resistance to a multitude of antibiotics. Using a combination of free and directed Molecular Dynamics (MD) simulations, we analyzed the interactions of MtrD w… Show more

“…These peptides likely enter gonococci and bind to this efflux pump and, as a consequence, decrease efflux activity. Advances in determining the structure of MtrD complexed with antibiotics in the proximal or deep binding pockets could facilitate structure-based approaches to develop specific, safe inhibitors that act at designated sites within this transporter [72][73][74][75]. With specific regard to gene regulation, the results of Chen et al [65] showing that genetic dampening of mtrCDE can increase PEN susceptibility provide a reason to use an RNA silencing approach to enhance antibiotic susceptibility.…”
