Transport of Pru p 3 across gastrointestinal epithelium – an essential step towards the induction of food allergy?

Abstract: The capacity of food allergens to cross the intestinal monolayer could explain their high allergenic capacity and its fast diffusion through the body associating to severe symptoms.

“…For example, the formation of lipid rafts and caveolas that allow endocytosis in epithelial cells of Prup p3, an nSLTP from peach. In addition, the reduction of Th2 cytokines of hypoallergenic peach nSLTP, compared to Prup p3 is associated with a lower transport capacity of the former (77). Der p2, may be internalized by the human airway epithelium and potentiate the airway inflammation because of its similar structure and function with the myeloid 2 differentiation protein (MD2), a TLR receptor signaling protein (78).…”

“…It has been speculated that the antimicrobial activity of nsLTPs could result from the interaction and permeabilization of biological membranes 66 . Tordesillas et al 14 showed that the nsLTP from peach, Pru p 3, was internalized by Caco 2 cells through an endocytic pathway involving lipid rafts and caveolar endocytosis. The authors further reported that the lower transport rate of a hypoallergenic peach nsLTP was associated with a significantly lower expression of T H 2-related cytokines compared with Pru p 3 (Table I).…”

“…Total RNA was extracted from the tissues following the method published (Tordesillas et al 2013). Gel electrophoresis (1% agarose) and spectrophotometry (260/280 nm) were used to determine RNA quality and concentration.…”

“…Finally, 10 ng of cDNA per well were added, and the final concentration of the primers was 1.5 uM. The amount of Pru p 3, STR, and S4D mRNA expression was normalized using the endogenous control 18S, and relative quantification was performed using the comparative threshold cycle method (2-AACt), as previously described (Tordesillas et al 2013). Changes in gene expression were calculated with reference to anthesis (7 dpa).…”
