Transporter protein and drug-conjugated gold nanoparticles capable of bypassing the blood-brain barrier

Abstract: Drug delivery to the central nervous system (CNS) is challenging due to the inability of many drugs to cross the blood-brain barrier (BBB). Here, we show that wheat germ agglutinin horse radish peroxidase (WGA-HRP) chemically conjugated to gold nanoparticles (AuNPs) can be transported to the spinal cord and brainstem following intramuscular injection into the diaphragm of rats. We synthesized and determined the size and chemical composition of a three-part nanoconjugate consisting of WGA-HRP, AuNPs, and drugs … Show more

“…Indeed, no brain changes were noted with H & E staining and GO deposition was not observed. These observations differ from studies of gold nanoparticles which can pass through the blood brain barrier [34]. Thus, GO may not be toxic to the central nervous system.…”

“…AuNPs have low toxicity and also easily cross the BBB [111]. For example, a wheat germ agglutinin horseradish peroxidase (WGA-HRP) conjugated AuNPs has been shown to be able to cross the BBB through intramuscular injection in the diaphragm of rats [112]. They are also used for imaging as X-ray contrast agents for computer tomography (CT) [113].…”
