Triclosan inhibits the activation of human periodontal ligament fibroblasts induced by lipopolysaccharide from Porphyromonas gingivalis

Abstract: Periodontitis is a highly prevalent, chronic, non-specific, and immunologically devastating disease of periodontal tissues, caused by microbial infection. This study aims to examine the efficacy and protective mechanism of triclosan (TCS), a bisphenolic, non-cationic component of oral care products, against periodontal inflammation induced by lipopolysaccharide purified from Porphyromonas gingivalis (LPS-PG). TCS markedly downregulated interleukin-6 (IL-6), IL-8, and IL-15 in human perio… Show more

“…Many investigators have reported that Pg -LPS induces the production of pro-inflammatory cytokines, i.e., IL-1, IL-6, IL-8, and tumor necrosis factor alpha (TNF-α), via the NF-κB pathway in HGFs [ 25 , 26 , 27 ]. HPLFs also perform a similar function to that of HGFs [ 28 , 29 ]. Furthermore, HPLFs share some of the properties of both osteoblasts and cementoblasts [ 30 ].…”

“…A recent study on rats demonstrated the therapeutic effectiveness of TCS and an anti-inflammatory flurbiprofen-loaded nanogels system in ligature-induced experimental periodontitis in rats [ 97 ]. TCS activity in periodontitis is related to the activation of human periodontal ligament fibroblasts that are induced by lipopolysaccharide from Porphyromonas gingivalis [ 98 ]. Recently, the link between TCS and chitosan, a natural polymer that may act as a drug delivery agent and exerts antibacterial and anti-inflammatory activities [ 99 ], has been investigated in the prevention and/or treatment of inflammation in periodontal diseases.…”
