“…Murine mammary carcinoma cell line EMT-6 and human colorectal adenocarcinoma cell line SW480, purchased from American Type Culture Collection (ATCC, Manassas, VA), were cultured in DMEM supplemented with 10% of fetal bovine serum and penicillin (100 U/mL), streptomycin (100 mg/mL), amphotericin B (250 ng/mL) and placed in CO 2 incubator with 5% of CO 2 at 37 C. Cells were seeded in a 96-well microplate at 3.125 Â 10 4 cells/mL and treated with compounds (10, 30 and 100 mM) for 48 h. Doxorubicin, etoposide, 5-fluorouracil and methotrexate (10 mM) were used as anti-cancer references. The anti-proliferative activity of compounds and drug references were determined using XTT assay 30,31 . This assay is based on the conversion of the water-soluble XTT (sodium 2,3,-bis(2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino)-carbonyl]-2H-tetrazolium) inner salt) reagent, in the presence of the electron-coupling reagent PMS (phenazine methylsulfate) to an orange formazan product by metabolically active cells.…”
