Tumor Cell Adhesion to Endothelial Cells: Endothelial Leukocyte Adhesion Molecule-1 as an Inducible Adhesive Receptor Specific for Colon Carcinoma Cells

Abstract: Activation of endothelial cells by the two inflammatory mediators interleukin-1 (IL-1) and tumor necrosis factor strongly increases tumor cell adhesion. We describe antibody inhibition studies showing that the endothelial leukocyte adhesion molecule-1 (ELAM-1), a cell-surface glycoprotein selectively expressed by cytokine-activated endothelial cells and responsible for neutrophil adhesion, is the major, if not the only, mediator of colon carcinoma cell adhesion to activated endothelial cells. Among the differe… Show more

“…Interestingly, elevated levels of sVCAM-1 were noted in several human malignancies such as in the ovary and breast, outside of the head and neck region (Banks et al, 1993). Expression of E-selectin is endothelium-specific (Bevilacqua and Nelson, 1993), and E-selectin-mediated adhesion of colon carcinoma and head and neck squamous cell carcinoma cells to endothelium is suggested to be associated with metastasis (Lauri et al, 1991;Wenzel et al, 1995). Soluble E-selectin is evaluated as a marker for endothelial damage after activation by cytokines (Gearing and Newman, 1993), the levels are higher in ovarian, breast and gastrointestinal cancers (Banks et al, 1993).…”

“…However, evidence for other selectins and tumor types is less convincing (Sass, 1998;Krause and Turner, 1999). Initially, it was shown that E-selectin supported adhesion of colon adenocarcinoma cells (Lauri et al, 1991), and it became apparent that these cells express sLeX and sLeA, which are both ligands for E-selectin (Takada et al, 1991;Majuri et al, 1992). SLeX and sLeA have been detected in cutaneous SCC with accompanying E-selectin induction in stroma around and within tumors (Groves et al, 1993).…”

“…VCAM-1 is also induced on endothelial cells, mediating adhesion of lymphocytes and monocytes, but additionally is present on lymphoid dendritic cells, some tissue macrophages and renal parietal epithelium (Rice et al, 1991). Possible role for these molecules in metastasis are suggested by the reports of VCAM-mediated adhesion of melanoma cells (Rice & Bevilacqua 1989) and E-selectin-mediated adhesion of colon carcinoma cells (Lauri et al, 1991) to endothelium. Soluble variants of these molecules in the circulation have recently been reported .…”
