Tumor Necrosis Factor-α- or Lipopolysaccharide-induced Expression of the Murine P-selectin Gene in Endothelial Cells Involves Novel κB Sites and a Variant Activating Transcription Factor/cAMP Response Element

Abstract: Tumor necrosis factor-␣ (TNF-␣) or lipopolysaccharide (LPS) increases expression of the P-selectin gene in murine, but not in human, endothelial cells. These mediators augment expression of a reporter gene driven by the murine, but not the human, P-selectin promoter in transfected endothelial cells. The regions from ؊593 to ؊474 and from ؊229 to ؊13 in the murine P-selectin promoter are required for TNF-␣ or LPS to stimulate reporter gene expression. Within these regions, we identified two tandem B elements, a… Show more

“…14 Because IL-6, MCP-1, and P-selectin gene promoters contain functional NF-kB binding sites essential for their induction in response to inflammatory stimuli, 15,16 we further assessed NF-kB activation using the electrophoretic mobility shift assay. The NF-kB DNA binding activity from nuclear extracts of mouse renal kidney cells infected with lentivirus containing miR-494 was markedly increased compared with the NF-kB DNA binding activity of control cells not expressing miR-494 ( Figure 5D, upper panel, lane 5 versus lane 4).…”

“…22 Because the IL-6 and P-selectin gene promoters contain functional NF-B-binding sites essential for their induction in response to inflammatory stimuli, 23,24 we further assessed NF-B activation by electrophoretic mobility shift assay (EMSA). Renal I/R injury caused increased NF-B DNA-binding activity in nuclear extracts from WT kidneys, which was further markedly increased in ATF3-KO kidneys ( Figure 5B, top).…”
