Tumorhead distribution to cytoplasmic membrane of neural plate cells is positively regulated by Xenopus p21-activated kinase 1 (X-PAK1)

Abstract: Tumorhead (TH) regulates neural plate cell proliferation during Xenopus early development, and gain or loss of function prevents neural differentiation. TH shuttles between the nuclear and cytoplasmic/cortical cell compartments in embryonic cells. In this study, we show that subcellular distribution of TH is important for its functions. Targeting TH to the cell cortex/membrane potentiates a TH gain of function phenotype and results in neural plate expansion and inhibition of neuronal differentiation. We have f… Show more

“…3B). This was further supported by expression of a mutant of X-PAK1 (KR-PAK1), which acts as a dominant-negative kinase and blocks endogenous X-PAK1 activation (Wu et al, 2007). Expression of KR-PAK1 inhibited CNC cell migration.…”

“…Zebrafish Dnd1 was shown to block the 430/427/302 family of miRNAs in the germ plasm that target repression of nanos and TDRD7 (Tudor domain-containing 7) (BehmAnsmant et al 2006;Wu et al 2007). In Xenopus, miRNAs mediate the rapid deadenylation of maternal mRNAs after MBT as well.…”

“…Ectopic overexpression of Tumorhead mutant localizes to the plasma membrane; this results in neural plate expansion and inhibition of neuronal differentiation, which leads to embryonic lethality. xPAK1 mediates the phosphorylation of Tumorhead and enhances its binding to the apical cortex and lateral cell membrane of neural plate epithelial cells, resulting in neural plate expansion and inhibition of neuronal differentiation; organisms without xPAK1 activity show loss of the Tumorhead phenotype [Wu et al, 2007].…”
