Two enhancer domains control early aspects of the complex expression pattern of Msx1

Abstract: To understand how the complex embryonic expression pattern of the Msx1 gene is produced a transgenic analysis of 13 kb of DNA around the Msx1 locus was carried out. Most of the extensive expression pattern of the Msx1 gene was reproduced in transgenics using the LacZ gene fused to 5 kb of Msx1 5' flanking DNA. Two enhancer domains were identified which produced this pattern. The distal element produced expression in the first arch and the nasal epithelium and was restricted to 240 bp. However, the proximal ele… Show more

“…In mice, an AP-2 homolog is expressed in the urogenital epithelium (albeit in both sexes) and at least one AP-2 homolog shows sexually dimorphic expression (albeit in the brain) (Coelho et al, 2005). The mouse Dr homolog Msx1 is expressed in the genital ridge (MacKenzie et al, 1997) and Msx2 functions in female reproductive tract development (Yin et al, 2006). In chick embryos, Msx1 and Msx2 are expressed male specifically in the Müllerian ducts (Ha et al, 2008).…”

“…A transgene containing 5 kb upstream fragment of Msx1 gene (MacKenzie et al, 1997;Takahashi et al, 1997), EGFP cDNA and SV40 polyadenylation sequence was used to generate pMsx1-EGFP mice. Mice were analyzed at E10.0 (n=5), E10.5 (n=4) and E11.5 (n=4).…”
