Type 1 Allergy-Induced Endolymphatic Hydrops and the Suppressive Effect of H1-Receptor Antagonist (Olopatadine Hydrochloride)

Abstract: The systemic sensitization with DNP-As produced allergy-induced experimental EH by type 1 hypersensitivity allergic reaction, and the development of this EH was prevented by histamine H(1)-receptor antagonists.

“…Although the immunological basis for the allergic origin of MD is not well understood, early studies in the field of otolaryngology demonstrate amelioration of vertigo, tinnitus, and hearing loss in response to the desensitization of patients to inhalant allergens and elimination diet containing food allergens (11). Animals actively sensitized with a specific Ag and subsequently exposed to the same develop endolymphatic hydrops in the inner ear (12). A clinical report has described the relationship between endolymphatic hydrops and an allergic response, which can be triggered by a food allergenIgE Ab reaction (13).…”

“…Although the notion of immune-mediated inflammation in idiopathic, sensorineural hearing loss has persisted for over six decades 16 , little progress has since been made on the nature of the participant cellular components and the mechanism through which they interact to bring about inflammatory reactions. That said, available evidences have pointed to ELS as a key immunologic interface 17 , 18 . The labyrinthine sac is believed to harbour inner ear antigens of various molecular weights 19 , which can invoke autoimmune response (type II hypersensitivity) from the host defense 20 .…”

“…Immediate type of food allergy is mediated by type I hypersensitivity reaction, involving mast cells and food-specific IgE antibodies. In animal studies, Meneire's Disease could be induced by type 1 hypersensitivity which in turn, could be prevented by histamine H(1)-receptor antagonists [35]. Translated into clinical practice, a significant percentage of patients with Meniere's disease and allergy showed improvement in their symptoms of tinnitus and vertigo when receiving specific allergy therapy [34].…”
