Type-specific identification of influenza viruses A, B and C by the polymerase chain reaction

Abstract: The aim of this study was to develop a polymerase chain reaction for specific detection of influenza A, B, and C RNA genomes. Three primer sets were selected from conserved regions of the genome coding for the non-structural proteins and were tested on 61 influenza A (22 H1N1, 9 H2N2, and 30 H3N2), 11 influenza B, and three influenza C isolates. Specific amplified products were obtained with all these strains after electrophoresis on a 2% agarose gel. The specificity of the reaction was increased by hybridizat… Show more

“…A transcrição reversa foi realizada com o kit High Capacity cDNA Archive (Applied Biosystems), conforme as instruções do fabricante. Para a amplificação das amostras, foi utilizado o método de reação em cadeia da polimerase (polymerase chain reaction, PCR) com primers específicos para a detecção de VSRH, MPVh, IA, IB, VPI-1, VPI-2, VPI-3 e picornavírus [11][12][13][14] .…”

“…The two hMPV RT-PCRs were integrated in the existing in-house RT-multiplex PCR as follows: primers for the hMPV N gene were combined with those for RSV, influenza viruses A and B (block 1), and primers for HMPV L gene were combined with those for adenoviruses and parainfluenza type 1 and type 3 viruses (block 2) [6,13,20,26,38]. RNA of RNA viruses was transcribed to cDNA before PCR was performed.…”

“…There have been some reports on the use of the RT-PCR method to detect the influenza C virus (3,4,24), but there have been few reports on its actual use in the surveillance of this virus. In 2004, more than 70 strains of influenza C virus were isolated in Yamagata and Miyagi Prefectures, and we wanted to learn whether an epidemic of influenza C had occurred on a national scale.…”
