Type VI secretion system sheaths as nanoparticles for antigen display

Abstract: The bacterial type 6 secretion system (T6SS) is a dynamic apparatus that translocates proteins between cells by a mechanism analogous to phage tail contraction. T6SS sheaths are cytoplasmic tubular structures composed of stable VipA-VipB (named for ClpV-interacting protein A and B) heterodimers. Here, the structure of the VipA/B sheath was exploited to generate immunogenic multivalent particles for vaccine delivery. Sheaths composed of VipB and VipA fused to an antigen of interest were purified from Vibrio cho… Show more

“…First, compared to that of soluble antigens, the extremely high surface area to volume ratio of particulate adjuvants provides multiple attachment sites for the antigens or other stimuli to be presented in a multivalent manner, which is important for APC activation . For example, Del and coworkers presented evidence that the T6SS antigen delivery system shows the potential to deliver more than one antigen simultaneously into the same APC, thus effectively promoting APC maturation . Second, as multi‐payload carriers, particulate adjuvants can also load many immunostimulatory materials, such as toll‐like receptor (TLR) agonists, generating hybrid supramolecules to activate APCs .…”
