Unraveling the Role of Angiogenesis in Cancer Ecosystems

Abstract: Activation of the tumor and stromal cell-driven angiogenic program is one of the first requirements in the tumor ecosystem for growth and dissemination. The understanding of the dynamic angiogenic tumor ecosystem has rapidly evolved over the last decades. Beginning with the canonical sprouting angiogenesis, followed by vasculogenesis and intussusception, and finishing with vasculogenic mimicry, the need for different neovascularization mechanisms is further explored. In addition, an overview of the orchestrati… Show more

“…Next to common targets for tumor therapy, such as cell cycle inhibitors or proapoptotic agents, there are often angiogenesis modulators, especially in combined treatments [13]. The solid tumors, in fact, require an increased blood supply to support their growth, thus, angiogenesis is critical for the initiation, growth, and metastasis of these tumors [14].…”

“…This ubiquitous phenomenon in health and disease of higher organisms [1], plays a crucial role in the natural processes of organ growth and repair [2], wound healing [3], or inflammation [4]. Angiogenesis imbalance contributes to numerous malignant, inflammatory, ischaemic, infectious, and immune diseases [2,5], such as cancer [6][7][8][9][10], rheumatoid arthritis [11], neovascular age-related macular degeneration [12], endometriosis [13,14], and diabetes [15].…”
